چکیده

Abstract Lipid nanoparticles (LNP) are essential components of messenger RNA (mRNA) vaccines that have been a cornerstone the COVID-19 public health response. These LNPs used for cell entry and protection mRNA. Others shown LNP BNT162b2 vaccine is able to augment immune response against target. However, it unknown how this stimulates innate cells such as monocytes participate in engagement adaptive We THP-1 monocyte reporter line nuclear factor kappa B (NF-κB) interferon regulatory (IRF) activation understand if empty could stimulate comparison with toll-like receptor (TLR) agonists. With line, we show BNT162b2’s initiate NF-κB, but not IRF dose-dependent manner. The NF-κB was similar low doses R848, TLR 7/8 agonist, well LPS, TLR4 agonist. Next, evaluated NF-kB absence MyD88 or TRIF using knockout lines. demonstrate reduced both In comparison, LPS primarily stimulated through MyD88, R848 relies on pathways. Our data can key pathways suggesting master regulator may induced by current mRNA platform. Moreover, our suggest use signaling cascades induce activation. Understanding mechanisms behind immunostimulatory capacity shed light modulating these enhance design. Defense Health Program, HU00012120094

An intact canonical NF-κB pathway is required for inflammatory gene expression in response to hypoxia.

Hypoxia is a feature of the microenvironment in a number of chronic inflammatory conditions due to increased metabolic activity and disrupted perfusion at the inflamed site. Hypoxia contributes to inflammation through the regulation of gene expression via key oxygen-sensitive transcriptional regulators including the hypoxia-inducible factor (HIF) and NF-κB. Recent studies have revealed a high d...

The Transcriptional Specificity of NF-κB Dimers Is Coded within the κB DNA Response Elements

Nuclear factor κB (NF-κB) regulates gene expression by binding to specific DNA elements, known collectively as κB sites, that are contained within the promoters/enhancers of target genes. We found that the identity of the central base pair (bp) of κB sites profoundly affects the transcriptional activity of NF-κB dimers. RelA dimers prefer an A/T bp at this position for optimal transcriptional a...

NF-κB regulation: the nuclear response

Nuclear factor B (NFB) is an inducible transcription factor that tightly regulates the expression of a large cohort of genes. As a key component of the cellular machinery NFB is involved in a wide range of biological processes including innate and adaptive immunity, inflammation, cellular stress responses, cell adhesion, apoptosis and proliferation. Appropriate regulation of NFB is critical for...

The metal nanoparticle-induced inflammatory response is regulated by SIRT1 through NF-κB deacetylation in aseptic loosening

Aseptic loosening is the most common cause of total hip arthroplasty (THA) failure, and osteolysis induced by wear particles plays a major role in aseptic loosening. Various pathways in multiple cell types contribute to the pathogenesis of osteolysis, but the role of Sirtuin 1 (SIRT1), which can regulate inflammatory responses through its deacetylation, has never been investigated. We hypothesi...

Contracting C2C12 myotubes release CCL2 in an NF-κB-dependent manner to induce monocyte chemoattraction.

Muscle inflammation following exercise is characterized by expression of inflammatory cytokines and chemokines. Exercise also increases muscle macrophages derived from circulating monocytes. However, it is unknown whether muscle cells themselves attract circulating monocytes, or what is the underlying mechanism. We used an in vitro system of electrical stimulation (ES) causing C2C12 myotube con...
